A study investigating an ethyl acetate extract of Amphimedon chloros reported 14 identified compounds via LC–MS profiling, including zamamidine (11.4%) and keramamine (10.2%).[3]
In vitro assays used MDA-MB-231 breast cancer cells with human periodontal ligament fibroblasts as a non-cancer control. The extract showed cytotoxicity against MDA-MB-231 with an IC₅₀ of 3.0 µg/mL, while fibroblast viability was largely unaffected in the 50–200 µg/mL range tested.[3]
The same work reported gene-expression changes in MDA-MB-231 consistent with increased apoptosis/cell-cycle regulation: Bax, p21, and caspase-3 upregulated, with Bcl2 and Cyclin D (Cyclin D1) downregulated, and pathway-level discussion involving PI3K/Akt and MAPK signaling as potential upstream regulators (within the study’s experimental context).[3]
